This is a reality for AbbVie's Experiential Interns.Larger doses are more commonly needed for pipeline assets, both for new indications and life cycle management. The volumes needed to achieve this dose by using a standard concentration is burdensome to the patient and can cause manufacturing troubles. Developing such high concentrations requires a new strategy to overcome solubility and viscosity limitations. One way to solve this problem is by suspending powder in non-aqueous vehicles. To date, the combination of TFF (for UF/DF) and Spray Drying has shown much promise, though there is room to optimize the process and develop deeper understanding of these technologies when applied to pharmaceutically relevant proteins.The goal of this Intern Project is to apply fundamental principles to both TFF and Spray Drying to gain a deeper understanding of the process and impact of formulation. This level of understanding will allow the team to optimize the formulations and processes for ultra-high concentration liquid formulations and future assets. Further, an understanding of these process steps can be used in non-high concentration formulations, since TFF and Spray Drying are common and scalable processes.KEY RESPONSIBILITIES INCLUDE:

• Develop deep understanding of protein processing techniques
• Design and perform process development-related experiments, e.g., Design of Experiments
• Run various processes, including Tangential Flow Filtration and Spray Drying
• Characterize samples from different steps by various analytical techniques, including: Size Exclusion Chromatography (SEC), Protein Concentration (Solo VPE). Imaged Capillary Isoelectric Focusing (icIEF), Particle Size Measurement (Laser Diffraction), Scanning Electron Microscopy (SEM)
• Communicate results and conclusions to key stakeholdersMINIMUM QUALIFICATIONS
  • Currently enrolled in university, pursuing a Bachelor's, Master's, or PhD in Chemical Engineering, Material Science, Physics, or other related field
  • Must be enrolled in university during the internship period
  • Expected graduation date between August 2023 - August 2026
  • Effective communication skills
  • Strong problem-solving skillsPREFERRED QUALIFICATIONS
    • Exposure to engineering or material science related concepts
    • Previous experience with laboratory work
    • Previous experience with protein characterization
